Determination of amyloid β (Aβ) isoforms and in particular the proportion of the Aβ 1-42 isoform in cerebrospinal fluid (CSF) of patients suspected of Alzheimer's disease might help in early diagnosis and treatment of that illness. Due to the low concentration of Aβ peptides in biological fluids, a preconcentration step prior to the detection step is often necessary. This study utilized on-chip immunoprecipitation, known as micro-immunoprecipitation (μIP). The technique uses an immunosorbent (IS) consisting of magnetic beads coated with specific anti-Aβ antibodies organized into an affinity microcolumn by a magnetic field. Our goal was to thoroughly describe the critical steps in developing the IS, such as selecting the proper beads and anti-Aβ antibodies, as well as optimizing the immobilization technique and μIP protocol. The latter includes selecting optimal elution conditions. Furthermore, we demonstrate the efficiency of anti-Aβ IS for μIP and specific capture of 5 Aβ peptides under optimized conditions using various subsequent analytical methods, including matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS), capillary electrophoresis, microchip electrophoresis, and immunoblotting. The jump throw is one of the most applied techniques in handball. Therefore, analyzing its underlying performance factors is important since they may provide a basis to adjust training interventions. Currently, the coach decides what needs to be improved in order to enhance the overall performance of an athlete. The aim of this study is to break down the performance factors in jump throws and to assign athletes to subgroups. This classification should allow athletes to benefit from the resulting group-specific training contents and may support a coach in deciding about the next steps in his training program. A total of 113 male handball players (age 15 ± 0.38 years, height 184.07 ± 7.1?cm, weight 74.83 ± 9.31?kg) participated in this study. From each athlete the standing reach height (SRH), the jumping reach height achieved with a countermovement jump (JRH), the maximal ball height achieved with a handball-specific single-legged jump and straight arm (BHJ), and the ball release height achieved in a jump throw (BHT) were measured. Performance factors were derived as differences between these measurements (JRH ? SRH = UJA for unspecific jump ability, BHJ ? JRH = SJT for specific jump technique, BHT ? BHJ = STT for specific throwing technique). A hierarchical cluster analysis was used to find groups of athletes based on the derived and scaled difference measures (UJA, SJT, STT). The dendrogram shows a clear distinction of four groups of athletes. Average values within each cluster reveal (A) one group with high UJA values, (B) one with low UJA values, (C) one with high SJT but low STT, and (D) one with low SJT but high STT. These results may be interpreted with respect to consequences for training because group (B) seems to benefit most from a general strength training while the properties of group (A), (C), and (D) recommend a focus on technical aspects of performance. 相似文献

The authors examined the occupational satisfaction of 107 counselor educator mothers and found that work‐to‐family enrichment, support from colleagues, and number of children under age 6 were significant predictors of occupational satisfaction. The level of electoral mobilization, which is generally lowerin European elections as compared to first-order national elections,has been found to be lower among party adherents as well. The aim of this study was to determine external and internal loads on the knee joint during downhill walking with and without hiking poles. Kinematic, kinetic and electromyographic data were collected from eight males during downhill walking on a ramp declined at 25 degrees. Planar knee joint moments and forces were calculated using a quasi-static knee model. The results were analysed for an entire pole-cycle as well as differentiated between single and double support phases and between each step of a pole-cycle. Significant differences between downhill walking with and without hiking poles were observed for peak and average magnitudes of ground reaction force, knee joint moment, and tibiofemoral compressive and shear forces (12-25%). Similar reductions were found in patellofemoral compressive force, the quadriceps tendon force and the activity of the vastus lateralis; however, because of a high variability, these differences were not significant. The reductions seen during downhill walking with hiking poles compared with unsupported downhill walking were caused primarily by the forces applied to the hiking poles and by a change in posture to a more forward leaning position of the upper body, with the effect of reducing the knee moment arm. 相似文献

Lower extremity joint loading during walking is strongly affected by the steepness of the slope and might cause pain and injuries in lower extremity joint structures. One feasible measure to reduce joint loading is the reduction of walking speed. Positive effects have been shown for level walking, but not for graded walking or hiking conditions. The aim of the study was to quantify the effect of walking speed (separated into the two components, step length and cadence) on the joint power of the hip, knee and ankle and to determine the knee joint forces in uphill and downhill walking. Ten participants walked up and down a ramp with step lengths of 0.46, 0.575 and 0.69 m and cadences of 80, 100 and 120 steps per minute. The ramp was equipped with a force platform and the locomotion was filmed with a 60 Hz video camera. Loading of the lower extremity joints was determined using inverse dynamics. A two-dimensional knee model was used to calculate forces in the knee structures during the stance phase. Walking speed affected lower extremity joint loading substantially and significantly. Change of step length caused much greater loading changes for all joints compared with change of cadence; the effects were more distinct in downhill than in uphill walking. The results indicate that lower extremity joint loading can be effectively controlled by varying step length and cadence during graded uphill and downhill walking. Kinesio-tape® has been suggested to increase blood circulation and lymph flow and might influence the muscle's ability to maintain strength during fatigue. Therefore, the aim of this study was to investigate the influence of gluteal Kinesio-tape® on lower limb muscle strength in non-fatigued and fatigued conditions. A total of 10 male rugby union players performed 20-m sprint and vertical jump tests before and after a rugby-specific fatigue protocol. The 20-m sprint time was collected using light gates (SMARTSPEED). A 9-camera motion analysis system (VICON, 100 Hz) and a force plate (Kistler, 1000 Hz) measured the kinematics and kinetics during a counter movement jump and drop-jump. The effect of tape and fatigue on jump height, maximal vertical ground reaction force, reactivity strength index as well as lower limb joint work were analysed via a two-way analysis of variance. The fatigue protocol resulted in significantly decreased performance of sprint time, jump heights and alterations in joint work. No statistical differences were found between the taped and un-taped conditions in non-fatigued and fatigued situation as well as in the interaction with fatigue. Therefore, taping the gluteal muscle does not influence the leg explosive strength after fatiguing in healthy rugby players. 相似文献
